EFFECT OF INSULIN IN THE PRESENCE OF GLUCOSE AND AFTER DEPLETION OF GLUCOSE, GLUCOSE 6-PHOSPHATE AND GLYCOGEN By ERNST B. CHAIN and PETER M. SENDER*

چکیده
In the absence of glucose, insulin stimulated the incorporation of 14C-labelled amino acids into protein by perfused rat hearts that had been previously substantially depleted of endogenous glucose, glucose 6-phosphate and glycogen by substrate-free perfusion. This stimulation was also demonstrated in hearts perfused with buffer containing 2-deoxy-D-glucose, an inhibitor of glucose utilization. It is concluded that insulin exerts an effect on protein synthesis independent of its action on glucose metabolism. Streptozotocin-induced diabetes was found to have no effect either on 14C-labelled amino acid incorporation by the perfused heart or on the polyribosome profile and amino acidincorporating activity of polyribosomes prepared from the non-perfused hearts of these insulin-deficient rats, which show marked abnormalities in glucose metabolism. Protein synthesis was not diminished in the perfused hearts from rats treated with anti-insulin antiserum. The significance of these findings is discussed in relation to the reported effects of insulin deficiency on protein synthesis in skeletal muscle.

Succinic Acid Monoethyl Ester and Metformin Regulates Carbohydrate Metabolic Enzymes and Improves Glycemic Control in Streptozotocin-Nicotinamide Induced Type2 Diabetic Rats
Objective. Succnic acid mono ethyl ester (EMS) was recently proposed as an insulinotropic agent for the treatment of non-insulin dependent diabetes mellitus. In the present study the effect of EMS and Metformin on the activities of carbohydrate metabolic enzymes in streptozotocinnicotinamide induced type 2 diabeteic model was investigated. Methods. EMS were injected intraperitonially at doses 2...

Experimental fish were sprint trained by individual chasing for 30 s on alternate days for 9 weeks. Ten trained and 10 untrained animals were rapidly freezeclamped at rest and 0 ,1 , 3 and 6h after a 5-min chase. Swimming speed of 10 fish in each group was measured in a 2-min chase. Phosphocreatine (PCr), creatine, adenosine triphosphate (ATP), adenosine diphosphate (ADP), adenosine monophospha...
متن کاملControl of glycogen synthesis by glucose, glycogen, and insulin in cultured human muscle cells.
A key feature of type 2 diabetes is impairment in the stimulation of glycogen synthesis in skeletal muscle by insulin. Glycogen synthesis and the activity of the enzyme glycogen synthase (GS) have been studied in human myoblasts in culture under a variety of experimental conditions. Incubation in the absence of glucose for up to 6 h caused an approximately 50% decrease in glycogen content, whic...
متن کاملGlycogen depletion rather than glucose 6- P increments controls early glycogen recovery in human cultured muscle.
In glycogen-containing muscle, glycogenesis appears to be controlled by glucose 6-phosphate (6- P) provision, but after glycogen depletion, an autoinhibitory control of glycogen could be a determinant. We analyzed in cultured human muscle the contribution of glycogen depletion versus glucose 6- P in the control of glycogen recovery. Acute deglycogenation was achieved by engineering cells to ove...

We have examined the onset and duration of the inhibitory effect of an intravenous infusion of lipid/heparin on total body carbohydrate and fat oxidation (by indirect calorimetry) and on glucose disappearance (with 6,6 D2-glucose and gas chromatography-mass spectrometry) in healthy men during euglycemic hyperinsulinemia. Glycogen synthase activity and concentrations of acetyl-CoA, free CoA-SH, ...
